L-asparaginase exerts anti-leukemia activity

چکیده
This study aimed to investigate the effects of L-Asparaginase (L-ASP) on leukemia cell proliferation, cell cycle distribution and geminin expression. Jurkat cells were treated with different concentration of LASP for 24 h, 48 h or 72 h. Cell proliferation was analysed by the CCK-8 assay, cell cycle distribution was examined by flow cytometry, and mRNA and protein expression levels of geminin were assessed by PCR and Western blot analysis. The results showed that L-ASP inhibited the proliferation of Jurkat cells in a doseand time-dependent manner. In addition, L-ASP at 0.5-10 U/ml induced G1 phase arrest of Jurkat cells. Geminin mRNA and protein expression remarkably decreased in Jurkat cells treated with L-ASP at 1.0-10 U/ml. In conclusion, our study suggests that the anti-leukemia mechanism of LASP is related to the arrest of leukemia cells in G1 phase and the downregulation of geminin expression.

Isolation and identification of L-asparaginase producing actinomycetes from Persian Gulf
Introduction: L-asparaginase is an anti-neoblastic agent used in the chemotherapy of lymphoblastic leukemia. This enzyme is widely distributed among microorganisms, plants and animals, but microorganisms have proved to be a better alternative for L-asparaginase, thus facilitating its large-scale production. Actinomycetes are filamentous, Gram-positive bacteria widely distributed in the marine e...
متن کاملEffect of Conjugation of Activated Glutaraldehyde-Nanochitosan with L-Asparaginase as an Anti Cancer Enzyme on its Stability and Physicochemical Properties
Introduction: The bacterial Asparaginase is used in the treatment of asparagine-dependent tumors, particularly lymphatic sarcoma and acute lymphoblastic leukemia. However, the instability of the enzyme increases the number of injections that are accompanied by high immune responses. The aim of this study was to investigate the conjugation of L-asparaginase with nanochitosan glutaraldehyde (NCG)...

متن کاملL-Asparaginase-producing Rouxiella Species Isolation, Antileukemia Activity Evaluation, and Enzyme Production Optimization
Background: L-Asparaginase (L-Asp) is used as an efficient anti-cancer drug, especially for acute lymphoblastic leukemia (ALL). Currently, two bacterial asparaginase isoenzymes are used for cancer treatment. Therefore, this research focused on isolating native bacteria with the ability to produce L-Asp. Materials and methods: L-Asp producing bacteria were isolated from soil samples on 9K medi...
متن کاملMiniaturized fiber-optic biosensor to monitor asparagine in clinical samples
Asparagine biosensor was constructed by co-immobilized anti-leukemic enzyme (L-asparaginase) and absorptive pH sensitive indicator on to plastic chips of 5mm through hydro sol-gel approach. Asparagine is used as a biomarker for detection of acute lymphoblastic leukemia. In this research work asparagine biosensor was developed focusing on L-asparaginase activity as bioassay principle. L-asparagi...
